Cytotaxonomic diagnosis of Trichomycterus diabolus (Teleostei: Trichomycteridae) with comments about its evolutionary relationships with co-generic species Neotropical Ichthyology
Torres,Rodrigo A.; Oliveira,Claudio; Foresti,Fausto.
The karyotype and the Ag-NOR location of a sample of Trichomycterus diabolus collected in the córrego Hortelã (Botucatu, São Paulo, Brazil) are described. The species exhibited 2n=56 chromosomes (42 metacentrics, 12 submetacentrics and 2 subtelocentrics) and the nucleolus organizing region located near to the centromere on the long arm of the largest metacentric pair. The presence of 2n=56 chromosomes in T. diabolus is an interesting characteristic since until now all cis-Andean species karyotyped presented 2n=54 chromosomes while almost all trans-Andean species presented different diploid numbers. The possible origin of this unexpected karyotypic form is discussed.

Description of a new species of Moenkhausia (Characiformes: Characidae) from the upper Paraguay basin, Central Brazil, with comments on its phylogenetic relationships Neotropical Ichthyology
Britzke,Ricardo; Troy,Waldo P.; Oliveira,Claudio; Benine,Ricardo C..
ABSTRACT A new species of Moenkhausia is described from tributaries of the upper rio Sepotuba, Paraguay basin, Brazil. The new species is distinguished from its congeners by a combination of characters, including an inconspicuous oval-shaped vertically elongated humeral blotch, extending horizontally from third through five lateral-line scales, and vertically from third row above lateral line to first row below it, followed by a diffuse field of dark chromatophores in the flank, combined with a well-defined dark line at the base of the anal fin. Furthermore, the phylogenetic position of the new species is presented based on molecular data, showing a close relationship among species of Moenkhausia and Hemigrammus that have a well-defined dark line at the...

Description of a ZZ/ZW sex chromosome system in Thoracocharax cf. stellatus (Teleostei, Characiformes, Gasteropelecidae) Genet. Mol. Biol.
Carvalho,Margarida Lima; Oliveira,Claudio; Foresti,Fausto.
The family Gasteropelecidae is composed of three genera and eight species. This study shows that Thoracocharax cf. stellatus has 2n = 52 chromosomes for both sexes. The five males studied showed 8 metacentric, 16 submetacentric, 4 subtelocentric, and 24 acrocentric chromosomes; the seven females showed only one submetacentric chromosome, belonging to pair 11, and one extra acrocentric chromosome, smaller than all the other chromosomes, characterizing the presence of a ZZ/ZW sex chromosome system in this species. Nucleolus organizing regions (NORs) were detected on the short arms of the subtelocentric chromosome pair 13. Constitutive heterochromatin was identified at pericentromeric and terminal positions in almost all chromosomes. The W chromosome was...

Development and characterization of microsatellite loci of Microglanis cottoides (Siluriformes: Pseudopimelodidae) and cross-species amplification Neotropical Ichthyology
Souza-Shibatta,Lenice; Ferreira,Dhiego Gomes; Oliveira,Claudio; Almeida,Fernanda Simões de; Shibatta,Oscar Akio; Sofia,Silvia Helena.
Thirteen microsatellite loci were isolated and characterized in Microglanis cottoides. Of these, two were monomorphic and 11 were polymorphic. These polymorphic loci tested on 24 individuals from a wild population produced a total of 108 different alleles, with levels of variability high, ranging from 2 to 20, with an average of 8.3 alleles per locus. The observed and expected heterozygosity ranged from 0.125 to 0.958 and from 0.119 to 0.931, respectively. A high combined probability of paternity exclusion value and a low probability combined genetic identity value obtained show that the set of loci described herein displays good suitability for paternity studies and differentiation of M. cottoides. Additionally, all thirteen microsatellite primers...

Discrimination of Shark species by simple PCR of 5S rDNA repeats Genet. Mol. Biol.
Pinhal,Danillo; Gadig,Otto BF; Wasko,Adriane P; Oliveira,Claudio; Ron,Ernesto; Foresti,Fausto; Martins,Cesar.
Sharks are suffering from intensive exploitation by worldwide fisheries leading to a severe decline in several populations in the last decades. The lack of biological data on a species-specific basis, associated with a k-strategist life history make it difficult to correctly manage and conserve these animals. The aim of the present study was to develop a DNA-based procedure to discriminate shark species by means of a rapid, low cost and easily applicable PCR analysis based on 5S rDNA repeat units amplification, in order to contribute conservation management of these animals. The generated agarose electrophoresis band patterns allowed to unequivocally distinguish eight shark species. The data showed for the first time that a simple PCR is able to...

Distributions and phylogeographic data of rheophilic freshwater fishes provide evidences on the geographic extension of a central-brazilian amazonian palaeoplateau in the area of the present day Pantanal Wetland Neotropical Ichthyology
Ribeiro,Alexandre C.; Jacob,Rodrigo M.; Silva,Ronnayana R. S. R.; Lima,Flávio C. T.; Ferreira,Daniela C.; Ferreira,Katiane M.; Mariguela,Tatiane C.; Pereira,Luiz H. G.; Oliveira,Claudio.
The analysis of the distribution patterns presented by examples of freshwater fishes restricted to headwater habitat: the anostomid Leporinus octomaculatus, the characins Jubiaba acanthogaster, Oligosarcus perdido, Moenkhausia cosmops, Knodus chapadae, Planaltina sp., the loricariid Hypostomus cochliodon, and the auchenipterid Centromochlus sp. provided evidences of a relatively recent shared history between the highlands of the upper rio Paraguay and adjoining upland drainage basins. Restricted to headwater of the uplands in the upper rio Paraguay and adjoining basins, these species provide biological evidence of the former extension of the central Brazilian plateau before the origin of the Pantanal Wetland. Disjunction took place due to an ecological...

DNA barcode of Parodontidae species from the La Plata river basin - applying new data to clarify taxonomic problems Neotropical Ichthyology
Bellafronte,Elisangela; Mariguela,Tatiane Casagrande; Pereira,Luiz Henrique Garcia; Oliveira,Claudio; Moreira-Filho,Orlando.
In the past years, DNA barcoding has emerged as a quick, accurate and efficient tool to identify species. Considering the difficulty in identifying some Parodontidae species from the La Plata basin and the absence of molecular data for the group, we aimed to test the effectiveness of DNA barcoding and discuss the importance of using different approaches to solve taxonomic problems. Eight species were analyzed with partial sequences of Cytochrome c oxidase I. The mean intraspecific K2P genetic distance was 0.04% compared to 4.2% for mean interspecific K2P genetic distance. The analyses of distance showed two pairs of species with K2P genetic divergence lower than 2%, but enough to separate these species. Apareiodon sp. and A. ibitiensis, considered as the...

Evidence of an XX/XY sex chromosome system in the fish Dormitator maculatus (Teleostei, Eleotrididae) Genet. Mol. Biol.
Oliveira,Claudio; Toledo,Lurdes Foresti de Almeida.
The fish Dormitator maculatus has a chromosomes number of 2n = 46, females having a karyotype of 14 M, 28 SM, 2 ST and 2A and males 13 M, 28 SM, 3 ST and 2A. The presence of a heteromorphic pair in the males and a corresponding homomorphic pair in the females suggest the occurrence of an XX/XY sex chromosome system in D. maculatus. The putative X chromosome has a pericentromeric C-band positive segment and the putative Y chromosome a C-band positive short arm.

Extensive polymorphism and chromosomal characteristics of ribosomal DNA in the characid fish Triportheus venezuelensis (Characiformes, Characidae) Genet. Mol. Biol.
Nirchio,Mauro; Oliveira,Claudio; Ferreira,Irani Alves; Granado,Angel; Ron,Ernesto.
The karyotype and chromosomal characteristics of the characid fish Triportheus venezuelensis were investigated using differential staining techniques (C-banding, Ag-NOR staining) and fluorescent in situ hybridization (FISH) with an 18S rDNA probe. The diploid chromosome number (2n = 52), karyotype composition and sex chromosome determination system of the ZZ/ZW type were the same as previously described in other species of the genus Triportheus. However, extensive variation regarding nucleolus organizer regions (NOR) different from other species was observed. 18S rDNA sequences were distributed on nine chromosome pairs, but the number of chromosomes with Ag-NORs was usually lower, reaching a maximum of four chromosomes. When sequential staining experiments...

Genetic variability of two populations of Pseudoplatystoma reticulatum from the Upper Paraguay River Basin Genet. Mol. Biol.
Abreu,Marcia Matos de; Pereira,Luiz Henrique Garcia; Vila,Vilma Barretto; Foresti,Fausto; Oliveira,Claudio.
Catfishes of the genus Pseudoplatystoma are very important species due to both their high commercial value and their ecological role as voracious predators. They undertake lengthy migratory movements during their life-cycle, this including reproductive migration which occurs from October to December in the rainy season. In the present study, seven microsatellite loci were analyzed to access genetic variability in two samples of P. reticulatum from the Upper Paraguay Basin. The loci were highly polymorphic (mean = 7.28). According to all analysis, the two samples of P. reticulatum revealed pronounced genetic differentiation. Fst value was 0.2290, Rst value 0.1067 and AMOVA 22.90% (Fst) and 10.67% (Rst), all being highly significant (p < 0.001). The...

Ichthyofauna of streams of the Rio Sapucaí basin, upper Rio Paraná system, Minas Gerais, Brazil Biota Neotropica
Azevedo-Santos,Valter M.; Britski,Heraldo A.; Oliveira,Claudio; Benine,Ricardo C..
Abstract: The Rio Sapucaí basin, in Minas Gerais State, Brazil, is one of the many watersheds of the upper Rio Paraná system. Ichthyofauna surveys in this basin, in general, are scarce. In addition, small rivers and streams of the region have been targets of anthropogenic actions (e.g., pollution) - which suggest that more ichthyological studies must be performed within the watershed. In this study we provide a survey of species that occur within three streams of the lower Rio Sapucaí basin. Samples were collected in April, July, and November 2017 and in May 2018. Collections resulted in 349 individuals belonging to 28 species, five orders, and 12 families. Among our findings are three putatively undescribed species and the first record of Oligosarcus...

Identification and description of distinct B chromosomes in Cyphocharax modestus (Characiformes, Curimatidae) Genet. Mol. Biol.
Santos,Lessandra Viviane De Rosa; Foresti,Fausto; Martins,Cesar; Oliveira,Claudio; Wasko,Adriane Pinto.
Cytogenetic analyses were performed in Cyphocharax modestus, collected at Paranapanema River and Tietê River (São Paulo State, Brazil). A karyotype with 2n = 54 chromosomes was observed in the animals from both Brazilian freshwater river systems. One to four B chromosomes were also detected in individuals from the Paranapanema River, which represents the probable first report of more than a single supernumerary element in a species of the Curimatidae group. C-banding revealed centromeric and telomeric heterochromatin blocks in several chromosomes of the normal karyotype complement of C. modestus. Moreover, while some B chromosomes were characterized by the complete absence of C-bands, others were totally heterochromatic. Although there was a prevalence of...

Identities among actin-encoding cDNAs of the Nile tilapia (Oreochromis niloticus) and other eukaryote species revealed by nucleotide and amino acid sequence analyses Genet. Mol. Biol.
Poletto,Andréia B.; Wasko,Adriane P.; Oliveira,Claudio; Azevedo,Alexandre; Carvalho,Robson F.; Silva,Maeli Dal Pai; Foresti,Fausto; Martins,Cesar.
Actin-encoding cDNAs of Nile tilapia (Oreochromis niloticus) were isolated by RT-PCR using total RNA samples of different tissues and further characterized by nucleotide sequencing and in silico amino acid (aa) sequence analysis. Comparisons among the actin gene sequences of O. niloticus and those of other species evidenced that the isolated genes present a high similarity to other fish and other vertebrate actin genes. The highest nucleotide resemblance was observed between O. niloticus and O. mossambicus a-actin and b-actin genes. Analysis of the predicted aa sequences revealed two distinct types of cytoplasmic actins, one cardiac muscle actin type and one skeletal muscle actin type that were expressed in different tissues of Nile tilapia. The...

Interspecific chromosomal divergences in the genus Characidium (Teleostei: Characiformes: Crenuchidae) Neotropical Ichthyology
Alves,José Carlos Pansonato; Paiva,Luiz Ricardo de Souza; Oliveira,Claudio; Foresti,Fausto.
Karyotypes of seven fish species of the genus Characidium, three of them studied for the first time, were characterized using conventional cytogenetic techniques (Giemsa staining, Ag-NOR, and C-banding). All species presented a diploid number of 2n=50, with only metacentric and submetacentric chromosomes, as observed in all Characidium species studied. In two species cells with one to three B chromosomes were observed. All species analyzed have a single NOR-bearing chromosome pair with morphological differences among them. Characidium cf. zebra shows heterochromatic blocks restricted to the pericentromeric regions of all chromosomes denoting the absence of a sex chromosome system. On the other hand, the species Characidium lanei, C. pterostictum, C....

Karyological characterization of four Neotropical fish species of the genus Hisonotus (Teleostei, Loricariidae, Hypoptopomatinae) from distinct Brazilian river basins Genet. Mol. Biol.
Andreata,Artur Antonio; Oliveira,Claudio; Foresti,Fausto.
The karyotypes of four Hisonotus species (two provisionally-named species A and D, H. nigricauda, and H. leucofrenatus) were found to have the same diploid number of 2n = 54 and interstitial silver-staining nucleolus organizer regions (Ag-NORs) located on the long arm of the largest metacentric pair. The C-banding pattern appeared to be species-specific, with one group (H. nigricauda and the unnamed species A and D) being characterized by small amounts of positive C-banded segments and containing a sub-group (species A and D) identified by a large positive C-banded segment on a small metacentric chromosome pair. The second group contained different samples of H. leucofrenatus, characterized by a larger amount of C-band positive segments spread over several...

Karyotype characterization of Mugil incilis Hancock, 1830 (Mugiliformes: Mugilidae), including a description of an unusual co-localization of major and minor ribosomal genes in the family Neotropical Ichthyology
Hett,Anne Kathrin; Nirchio,Mauro; Oliveira,Claudio; Siccha,Zoila Raquel; Rossi,Anna Rita; Sola,Luciana.
This study reports the description of the karyotype of Mugil incilis from Venezuela. The chromosome complement is composed of 48 acrocentric chromosomes, which uniformly decrease in size. Therefore, the homologues can not be clearly identified, with the exception of one of the largest chromosome pairs, classified as number 1, whose homologues may show a subcentromeric secondary constriction, and of chromosome pair number 24, which is considerably smaller than the others. C-banding showed heterochromatic blocks at the centromeric/pericentromeric regions of all chromosomes, which were more conspicuous on chromosomes 1, given the C-positive signals include the secondary constrictions. AgNO3 and fluorescent in situ hybridization (FISH) with 45S rDNA...

Karyotype description and evidence of multiple sex chromosome system X1X1X2X2/X1X2Y in Potamotrygon aff. motoro and P. falkneri (Chondrichthyes: Potamotrygonidae) in the upper Paraná River basin, Brazil Neotropical Ichthyology
Cruz,Vanessa Paes da; Shimabukuro-Dias,Cristiane Kioko; Oliveira,Claudio; Foresti,Fausto.
Cytogenetic analysis of Potamotrygon aff. motoro and P. falkneri indicated the occurrence of an X1X1X2X2/X1X2 Y multiple sex chromosome system in both species, with 2n = 66 chromosomes for females and 2n = 65 chromosomes for males. The nucleolus organizer regions (NORs) identified using Ag-NOR technique showed that both species have multiple Ag-NORs (5 to 7 chromosomes stained). C-banding technique indicated the presence of heterochromatic blocks in the centromeric regions of almost all chromosomes in both species. Through this study there was evidence of heterogeneity in the karyotypes, which suggests that chromosomal rearrangements such as inversions and/or translocations occurred during the chromosomal evolution in two species of this genus.

Karyotype description of five species of Trichomycterus (Teleostei: Siluriformes: Trichomycteridae) Genet. Mol. Biol.
Sato,Luciana Ramos; Oliveira,Claudio; Foresti,Fausto.
Trichomycteridae is a family of small catfish which are widely distributed throughout Southern Central America and South America. The present study showed that the cis-Andean species Trichomycterus florensis, Trichomycterus sp. aff. Trichomycterus itatiyae, Trichomycterus reinhardti, Trichomycterus davisi and Trichomycterus auroguttatus had 2n = 54 chromosomes (42 metacentric, 10 submetacentric and 2 subtelocentric), with T. reinhardti, T. auroguttatus and T. sp. aff. T. itatiyae exhibiting only one chromosome pair with silver-stained nucleolus organizer regions (NORs). The cytogenetic data suggest the existence of at least two groups of species in the cis-Andean representatives of the genus Trichomycterus. In the first group the first metacentric pair is...
